qRTPCR Technique Overview

qRT-PCR (quantitative Reverse Transcription Polymerase Chain Reaction) is a cornerstone technique in molecular biology used to measure gene expression with high sensitivity and precision. Our video provides a clear, step-by-step breakdown of the process, from reverse transcription using reverse transcriptase—converting RNA into complementary DNA (cDNA)—to the thermal cycling stages of PCR: denaturation, annealing, and extension. We explain how SYBR Green dye binds to double-stranded DNA, allowing fluorescence to track DNA amplification in real time. The concept of Ct values and how they relate to initial RNA concentration is also covered, alongside the importance of avoiding common errors like incorrect threshold settings or degraded RNA.
